The aim of this book will be to discuss recent advances in the field of matrix metalloproteinase (MMP) research from a multidisciplinary perspective. Chapters will be written by leaders in the field of MMPs. The focus of the book will be on the challenges of understanding the mechanisms substrate degradation by MMPs. In addition, the book will focus on how these enzymes are able to degrade large, highly ordered substrates such as collagen. All of these topics will be considered with their relationship to disease progression including their role in cancer metastisis, rheumatoid arthritis and other inflammatory diseases. Recently there has been a surge of research in the field of MMPs. The field is undergoing a very exciting transformation. Therefore, an updated book focusing on the past and exciting future of the field is warranted. The book will start with an overview of the matrix metalloproteinases, followed by a focus on the history of the proteins and the development and subsequent failure of the small molecule inhibitors in clinical trials. Furthermore, work with TIMPS, the endogenous inhibitors of the MMPs will be reviewed. The introductory chapters will establish the foundation for the later chapters as they will discuss the recent progress on the design of different types of inhibitors including novel antibody based therapeutics. In this second section, there will be an emphasis on those laboratories that have combined novel methods to further the study of the MMPs. In the third section, the focus will be in-vivo work which will focus on cancer models, degradation of the exracellular matrix, as well as the MMPs involvement in other disease states. The final chapter will focus the future of the field, and to which topics members of the field will need to concentrate their future efforts.
